Growth hormone releasing peptide-2 (GHRP-2) is a synthetic analog of ghrelin, a gastrointestinal peptide that binds to the growth hormone secretagogue receptor (GHS-R). Ghrelin increases growth hormone release as well as hunger.1 In a similar way, GHRP-2 activates GHS-R triggering the release of GH. Both ghrelin and GHRP-2 also stimulate the release of prolactin, cortisol, and corticotropin from the anterior pituitary gland.2 GHRP-2 contains 6 amino acids and has a molecular weight of 818 kDa. Despite the different chemistry of natural ghrelin and GHRP-2, evidence strongly supports that they have the same biological actions as discussed above.3 GHRP-2 acts in two ways to boost natural growth hormone levels. First, GHRP-2 amplifies the endogenous signal for growth hormone secretion by interaction with GHS-R, resulting in substantial growth hormone release.3 In a lab experiment, GHRP-2 significantly increased the release of growth hormone in mice carrying a genetic mutation in natural growth hormone releasing hormone (GHRH).4 Second, it suppresses somatostatin, which inhibits growth hormone.5 Furthermore, GHRP-2 has been shown to work in conjunction with the growth hormone-releasing factor (GRF) to release growth hormone from the pituitary gland of rats.5 Animal and human studies show GHRP-2 increases food intake.3,6 In addition to this, GHRP-2 improves muscle growth7, immunity8, sleep quality9, and may protect the heart.10 Further, GHRP-2 increases the release of insulin-like growth factor I (IGF-I).11 A study found that intraperitoneal injection of GHRP-2 reduced tissue necrotic factor-alpha (TNF-α) in a rat model of liver damage, suggesting a role of GHRP-2 in inflammation.12 GHRP-2 is a useful reagent used in in vitro experiments with its multidimensional effects. GHRP-2 comes in a vial as a lyophilized powder (5 mg/vial). The vial content can be reconstituted with a convenient volume of sterile water and used in vitro studies.
